Synvisc Injection Main Ingredients and Device Properties

Synvisc injection is made up of hydrated hylan A and B polymers derived from rooster combs. These polymers are chemically cross-linked to improve viscosity and elasticity, resulting in a viscoelastic injectable gel that closely resembles natural synovial fluid. Each syringe contains hylan, along with sodium chloride, disodium hydrogen phosphate, sodium dihydrogen phosphate hydrate, and sterile water for injection.

Synvisc is supplied in a 2.25 mL glass syringe, containing a 2 mL dose of hylan G-F 20. The product also comes with traceability labels and a product pamphlet. 

Storage Conditions

Store in original packaging (protected from light) at room temperature below 86°F (30°C). Do not freeze.

Mode of Administration

Synvisc is administered through an intra-articular injection, typically once a week for three weeks, with each injection spaced one week apart. Prior to each injection, synovial fluid or joint effusion should be removed from the knee. An 18- to 22-gauge needle is used for this process. To minimise leakage during administration, the tip cap should be twisted before being removed, and the needle must be tightly secured on the syringe.

The entire 2 mL dose is injected into one knee only. Synvisc is intended solely for injection into the knee by a healthcare professional and has not been tested for pain relief in other joints.

Post-Treatment Care

After the injection, patients should avoid strenuous activities, including high-impact sports and prolonged weight-bearing activities, for about 48 hours. Patients should consult their physician before resuming these activities.

How Long Do Synvisc Effects Last?

The effects of Synvisc injections can last up to 6 months, providing significant pain relief and improved knee function.

Potential Side Effects

After receiving Synvisc injections, some patients may experience temporary pain, swelling, or fluid build-up (effusion) in the treated joint. Significant swelling can cause discomfort, and patients should consult a doctor if this occurs.

The most common side effects include pain, swelling, and joint effusion in the injected knee. Other possible reactions are joint stiffness, warmth, injection site pain, arthritis, and gait disturbance. Rest, ice, or pain relievers may help manage these symptoms.

In rare cases, allergic reactions may occur, with symptoms like rashes, itching, swelling of the face, fast heartbeat, nausea, and difficulty breathing. In very rare instances, joint infections or low platelet counts have been reported.

Contraindications for the Treatment

Synvisc should not be administered to patients with:

  • Known hypersensitivity or allergies to hyaluronan or sodium hyaluronate products
  • Knee joint infections or skin diseases at the injection site
